检测原理 | 本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性捕获抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的待测物质与捕获抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物(TMB),避光显色。颜色反应的深浅与样本中待测物质的浓度成正比。加入终止液终止反应,在450nm波长(参考波长570 - 630nm)测定吸光度值。 |

CDH1 分子靶点综述
CDH1,又名上皮细胞钙粘素和CD324,是一种钙依赖性的细胞粘附分子。它包括胞外结构域的5个重复钙粘蛋白、一个跨膜结构域和一个可结合p120-联蛋白和β-联蛋白的胞内结构域。成人组织的CDH1在上皮组织中表达,在那里它可不断地再生,它在细胞表面的半衰期是5个小时。CDH1功能或表达缺失与肿瘤的进展和转移有关,下调CDH1的表达可降低细胞在组织中的粘附强度,导致细胞运动增加。反过来,它可允许癌细胞穿过基底膜,浸入周围组织。病理学家通常用CDH1来诊断不同类型的乳腺癌。免疫组化研究表明,与浸润性导管癌相比,浸润性小叶癌的CDH1表达显著降低或绝大多数缺失。
