检测原理：本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人CDH1抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、检测样本和辣根过氧化物酶标记的检测抗体，经过孵育，样本中存在的CDH1与固相抗体和检测抗体结合。洗涤后，加入显色底物TMB，避光显色。颜色反应的深浅与样本中CDH1的浓度成正比。加入终止液终止反应，在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

CDH1 分子靶点综述

CDH1，又名上皮细胞钙粘素和CD324，是一种钙依赖性的细胞粘附分子。它包括胞外结构域的5个重复钙粘蛋白、一个跨膜结构域和一个可结合p120-联蛋白和β-联蛋白的胞内结构域。成人组织的CDH1在上皮组织中表达，在那里它可不断地再生，它在细胞表面的半衰期是5个小时。CDH1功能或表达缺失与肿瘤的进展和转移有关，下调CDH1的表达可降低细胞在组织中的粘附强度，导致细胞运动增加。反过来，它可允许癌细胞穿过基底膜，浸入周围组织。病理学家通常用CDH1来诊断不同类型的乳腺癌。免疫组化研究表明，与浸润性导管癌相比，浸润性小叶癌的CDH1表达显著降低或绝大多数缺失。
