检测原理 | 本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人THBS-1抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的THBS-1与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中THBS-1的浓度成正比。加入终止液终止反应,在450nm波长(参考波长570 - 630nm)测定吸光度值。 |

THBS1 分子靶点综述
血小板反应素-1(THBS-1)是血小板反应素家族成员,介导细胞-细胞和细胞-基质相互作用。该蛋白可与纤维蛋白原、纤维连接蛋白、层粘连蛋白、V型胶原和整合素α-V/β-1结合。它对血小板聚集、血管生成和肿瘤发生发挥作用。在正常组织中,它是新血管形成和肿瘤发生的天然抑制剂。THBS-1还可正向和负向调控内皮细胞黏附、运动和生长。通过阻断THBS-1与细胞表面受体(CD47)结合,正常组织几乎对肿瘤辐射治疗产生免疫,并有助肿瘤死亡。
