CDH1 分子靶点综述

CDH1，又名上皮细胞钙粘素和CD324，是一种钙依赖性的细胞粘附分子。它包括胞外结构域的5个重复钙粘蛋白、一个跨膜结构域和一个可结合p120-联蛋白和β-联蛋白的胞内结构域。成人组织的CDH1在上皮组织中表达，在那里它可不断地再生，它在细胞表面的半衰期是5个小时。CDH1功能或表达缺失与肿瘤的进展和转移有关，下调CDH1的表达可降低细胞在组织中的粘附强度，导致细胞运动增加。反过来，它可允许癌细胞穿过基底膜，浸入周围组织。病理学家通常用CDH1来诊断不同类型的乳腺癌。免疫组化研究表明，与浸润性导管癌相比，浸润性小叶癌的CDH1表达显著降低或绝大多数缺失。
