检测原理 | 本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗小鼠PDGF-BB抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的PDGF-BB与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中PDGF-BB的浓度成正比。加入终止液终止反应,在450nm波长(参考波长570 - 630nm)测定吸光度值。 |

血小板衍生因子(PDGF)是众多调节细胞生长和分裂的因子之一。从化学结构上来说,PDGF是一种由两条A链(-AA)或两条B链(-BB)或AB链组合(-AB)形成的二聚体糖蛋白。特别是,它在血管生成、已存在的血管组织生长中起着重要作用。不受控制的血管生成是肿瘤的一个特征。肝细胞和不吸收的破骨细胞表达PDGF-BB,产生成骨细胞和骨头,血小板、巨噬细胞和肥大细胞也能产生PDGF-BB。在损伤部位,PDGF-BB促进中性粒细胞和巨噬细胞浸润,发挥清创作用,促进成纤维细胞分泌新的细胞外基质,促进IGF-1介导再上皮化。
