KDR 分子靶点综述

血管内皮生长因子受体2(VEGFR2)也称为激酶插入区受体(KDR),是酪氨酸激酶受体(RTKs)III类亚家族成员。与VEGFR1相比,VEGFR2更广泛分布表达于说要血管衍生的内皮细胞。VEGFR2与VEGF-A(VEGF121、VEGF165、VEGF189和VEGF206剪切变体)、VEGF-C和VEGF-D结合。可溶性VEGFR1和VEGFR2在阻断VEGF诱导的细胞增殖和迁移方面差异非常显著。可溶性VEGFR2与可溶性VEGFR1相反,它无法与VEGF竞争,与表达VEGFR1和VEGFR2的内皮细胞结合。可溶性VEGFR2只能部分抑制细胞迁移,而可溶性VEGFR1几乎完全阻断VEGF诱导的细胞增殖和迁移。VEGF与可溶性VEGFR2而非VEGFR1的结合也依赖于肝素。VEGF/VEGFR2信号通路在肿瘤发生和其它病理性血管发生的疾病中具有重要作用。利用阻断抗体使功能性VEGFR2功能失活可破坏血管生成,阻止肿瘤细胞扩散。
