MMP3 分子靶点综述

基质金属蛋白酶3(MMP-3)，又名基质分解素-1，在人类中由MMP3基因编码。MMP3的表达主要在转录水平上调控，转录水平上的基因启动子对多种刺激物包括生长因子、细胞因子、促癌剂和原癌基因产物作出应答。MMP-3酶降解II、III、IV、IX和X型胶原蛋白、蛋白聚糖、纤维连接蛋白、层粘连蛋白和弹性蛋白。另外，MMP-3还可激活其它MMPs如MMP-1、MMP-7和MMP-9，因此MMP-3在结缔组织重塑中起关键作用。该酶参与伤口修复、动脉粥样硬化进展和肿瘤启动。MMP-3野生型和基因敲除的小鼠研究表明，MMP-3增加了外伤性损伤后的血脑屏障通透性。细胞核MMP-3可作为转录因子和蛋白酶发挥作用。
