检测原理：本试剂盒采用固相竞争酶联免疫吸附检测技术。特异性抗人ET-1抗体预包被在高亲和力的酶标板上。酶标板孔中加入生物素化的ET-1和未标记的ET-1或样本，竞争结合包被抗体上有限的结合位点。洗涤去除未结合的物质后，加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后，加入显色底物TMB，避光显色。颜色反应的深浅与样本中ET-1的浓度成反比。加入终止液终止反应，在450 nm波长(参考波长570 - 630 nm)测定吸光度值。

EDN1 分子靶点综述

内皮素-1(ET-1)是由21个氨基酸残基组成的多肽，是一种多效性分，它最初是212个氨基酸的前多肽原。血管内皮细胞是ET-1的丰富来源。内皮细胞中的ET-1的表达受多种因素的影响，包括机械刺激、多种激素和促炎性细胞因子。内皮素首先被鉴定为内皮素衍生的舒张因子，然后又被鉴定为内皮素衍生的收缩因子。ET-1刺激心肌收缩和心肌细胞生长，调节血管活性物质的释放，刺激平滑肌细胞有丝分裂。它也作为内皮细胞的促生因子，由下丘脑和垂体细胞调节其分泌。ET-1可能通过促进中性粒细胞的粘附和迁移以及刺激促炎细胞因子的产生来控制炎症反应。
