检测原理 | 本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人Bcl-2抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、检测样本和辣根过氧化物酶标记的检测抗体,经过孵育,样本中存在的Bcl-2与固相抗体结合和检测抗体结合。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中Bcl-2的浓度成正比。加入终止液终止反应,在450nm波长(参考波长570 - 630nm)测定吸光度值。 |

B细胞淋巴瘤2 (Bcl-2),是Bcl-2家族调节蛋白中的创始成员,可通过诱导凋亡(促凋亡)或者抑制凋亡(抗凋亡)来调节细胞凋亡。Bcl-2已明确是一种重要的抗凋亡蛋白,因此被归类为是一种致癌基因。Bcl-2调节线粒体动力学,并参与线粒体融合与分裂的调控。另外,在胰岛β细胞中,Bcl-2和Bcl-XL参与控制代谢活性和胰岛素分泌。 Bcl-2基因损伤会引发许多癌症,包括黑色素瘤、乳腺癌、前列腺癌、慢性淋巴细胞白血病和肺癌,还可能与精神分裂症和自身免疫性疾病有关。Bcl-2也是癌症治疗抵抗的一个原因。
