IGF1 分子靶点综述

胰岛素样生长因子1(IGF-1)，也称为生长调节素C，是一个在分子结构上类似于胰岛素的激素。IGF-1主要由肝脏产生，作为内分泌激素。同样地，靶组织亦能旁分泌/自分泌产生IGF-1。IGF-1是AKT信号转导途径中的一种有效天然激活剂、细胞生长和增殖的刺激子、细胞程序性死亡的有效抑制子。IGF-1最少可结合两种细胞表面受体：IGF-1受体(IGF1R)和胰岛素受体。IGF-1的产生受生长激素刺激，受营养不良、生长激素不敏感、缺少生长激素受体或者生长激素受体(SHP2和STAT5B)下游信号通路发生故障等方面所抑制。IGF-1在儿童生长过程中起重要作用，继而在成人合成代谢中有影响，且与老年化、神经病变、癌症以及中风相关。
