检测原理 | 本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人HGF抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的HGF与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中HGF的浓度成正比。加入终止液终止反应,在450nm波长(参考波长570 - 630nm)测定吸光度值。 |

肝细胞生长因子(HGF)是旁分泌性细胞生长、运动和形态发生的因子,由间充质细胞分泌产生,可作为大部分上皮来源细胞的多功能细胞因子。HGF以单链形式存在,是一个带有29个氨基酸信号序列和25个氨基酸前体序列的728个氨基酸残基前肽。HGF结合原癌基因c-Met受体后就会激活酪氨酸激酶信号级联,进而调节细胞的生长、运动和形态发生。它在血管生成、肿瘤发生和组织再生中起核心作用。目前正在测试人HGF质粒DNA治疗心肌细胞是否可作为冠状动脉疾病和心肌梗死后心脏损伤的一种潜在治疗。HGF也可作为基孔肯雅病毒诱导的关节痛的预后指示。高水平的HGF与与快速康复有关。
