Acts upstream of or within several processes, including positive regulation of T cell tolerance induction; regulation of T cell activation; and regulation of gene expression. Located in cytoplasm and nucleus. Is expressed in central nervous system; genitourinary system; hemolymphoid system; and retina. Used to study Wiskott-Aldrich syndrome and immune dysregulation-polyendocrinopathy-enteropathy-X-linked syndrome. 使用方法

取 100 μl 抗凝血或含有 1×10⁵ - 10⁸ 个单细胞的细胞悬液，根据实验方案和产品说明书，加入适量抗体进行表面染色，震荡混匀，于室温避光孵育适当时间。
将 Fixation/Permeabilization Concentrate (4×) (组分编号 ICC01)和 Fixation/Permeabilization Diluent(组分编号 ICD01)按 1：3 比例制备成 1×工作液。在流式管中染色，每个样本需要 1 ml。保存勿超过 1 天。用蒸馏水将 Permeabilization Buffer (10×) (组分编号 ICB01)稀释为 1×工作液。在流式管中染色，每个样本需要 8.5 ml。剩余溶液在 2 - 8℃最多保存 1 周。准备加入 1 ml
每管加入 1 ml Fixation/Permeabilization 工作液，涡旋震荡混匀。室温避光孵育 30 - 60 分钟。
无需洗涤，每管加入 2 ml 1× Permeabilization Buffer。室温下 300 - 400 × g 离心 5 分钟，弃去上清。
(可选)重复步骤 4。
用 100 μl 1× Permeabilization Buffer 重悬沉淀。通常洗涤后残留的液体量就已足够。
(可选)细胞悬液中加入 2 μl Anti-Mouse CD16/CD32, Purified (目录号 AM016)进行阻断，室温避光孵育 15 分钟。
无需洗涤，加入 5 μl Anti-Mouse Foxp3, PE 或 0.125 μg Mouse IgG1 Isotype Control, PE (目录号CMG104)。震荡混匀，室温避光孵育 15 分钟。
每管加入 2 ml 1× Permeabilization Buffer，室温下 300 - 400 × g 离心 5 分钟，弃去上清。
(可选)重复步骤 9。
每管加入 500 μl 1× Flow Cytometry Staining Buffer (目录号 S1001)重悬，上机检测。
