检测原理 | 本试剂盒采用固相竞争酶联免疫吸附检测技术。特异性抗小鼠/大鼠Insulin抗体预包被在高亲和力的酶标板上。酶标板孔中加入生物素化的Insulin和未标记的Insulin或样本,竞争结合包被抗体上有限的结合位点。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中Insulin的浓度成反比。加入终止液终止反应,在450nm波长(参考波长570 - 630nm)测定吸光度值。 |

INS 分子靶点综述
胰岛素是一种由胰岛β细胞和一些硬骨鱼布氏体分泌产生的肽类激素。它通过促进脂肪、肝脏和骨骼肌细胞吸收血液中的葡萄糖,从而对糖类、脂肪和蛋白质代谢起重要作用。这些组织吸收的葡萄糖转化为糖原或者脂肪(甘油三酯), 而肝脏是个例外,其吸收的葡萄糖可以转化为糖原和脂肪。肝脏中的葡萄糖产量受血液中高浓度胰岛素强烈抑制。血液中胰岛素浓度高时,它就是一种同化激素,可促进血液中的小分子转化为细胞内的大分子。血液中胰岛素浓度低时,它通过促进广泛的分解代谢而产生相反的效果。当胰岛β细胞被自身免疫过程破坏,胰岛素就不再合成或者分泌到血液中,进而导致 1 型糖尿病。遗传易感性和环境因素的综合影响会导致胰岛素抵抗,这就是 2 型糖尿病。另外,胰岛β细胞产生过量的胰岛素或反应性低血糖都与胰岛素瘤有关。
