酪氨酸蛋白激酶受体Tie1是一个单次跨膜的I型膜蛋白,属于蛋白激酶超家族、酪氨酸蛋白激酶家族和Tie亚家族。Tie1有三个EGF样结构域、三个纤连蛋白III型结构域、两个免疫球蛋白样C2型结构域和一个蛋白激酶结构域。Tie1是一个专门在内皮细胞中表达的细胞表面蛋白,然而,研究表明它同样也可在不成熟的造血细胞和血小板中表达。在血管发育的毛细管涌现阶段,Tie1对于微血管内皮细胞的存活和生长是必需的。Tie1通过p38依赖性机制上调细胞粘附分子VCAM-1、E-selectin和ICAM-1。Tie1的表达可增强单核细胞来源的免疫细胞粘附于内皮细胞。Tie1具有促炎症性效果,在内皮的炎症性疾病如动脉粥样硬化中起重要作用。
