TNFRSF9 分子靶点综述
4-1BB,也称为TNFRSF9或CD137,是肿瘤坏死因子超家族的受体。人4-1BB基因编码由255个氨基酸组成的I型跨膜蛋白,包含17个氨基酸的N端信号肽、169个氨基酸的胞外区、27个氨基酸的跨膜区和42个氨基酸的胞外区。重组人可溶性4-1BB是一个167氨基酸的多肽(17.7kDa),包括富含胱氨酸TNFR样的4-1BB胞外区。4-1BB主要表达于各种T细胞的表面,在B细胞、单核细胞和多种转化细胞系中也有表达。该蛋白有助于克隆增殖、存活和T细胞发育。它也能诱导外周单核细胞的增殖,增强由TCR/CD3触发活化而诱导的T细胞凋亡,同时调节CD28共刺激促进Th1细胞应答。该蛋白的表达是通过淋巴细胞活化诱导。TRAF转接蛋白可与该蛋白结合,并转导激活NF-κB的信号。它还参与高脂肪食源性代谢综合症中炎症的发展。
