CD6 分子靶点综述

分化簇6(CD6,亦称TP120)是一种单次跨膜的I型膜蛋白,其结构中含有三个SRCR结构域。该细胞表面糖蛋白主要表达于T细胞,可作为共刺激分子参与自身反应性免疫应答。CD6在胸腺细胞、成熟T细胞、B细胞亚群(B-1细胞)及部分脑细胞中均有表达。
研究已鉴定出CD6的配体CD166(曾用名:活化白细胞粘附分子ALCAM),该配体表达于活化的T细胞、B细胞、胸腺上皮细胞、角质形成细胞及类风湿性关节炎滑膜组织。CD6通过与活化白细胞粘附分子(CD166)结合,作为共刺激分子参与淋巴细胞活化与胸腺细胞发育过程。
在T淋巴细胞中,CD6部分与TCR/CD3复合体结合,并在成熟免疫突触(IS)中心与其共定位。胸腺发育过程中,CD6依赖性信号不仅促进胸腺细胞存活,还在人类和小鼠的选择过程中调控整体功能亲和力。
