检测原理 | 本试剂盒采用双抗体夹心酶联免疫吸附检测技术。特异性抗人IL-6Rα抗体预包被在高亲和力的酶标板上。酶标板孔中加入标准品、待测样本和生物素化的检测抗体,经过孵育,样本中存在的IL-6Rα与固相抗体和检测抗体结合。洗涤去除未结合的物质后,加入辣根过氧化物酶标记的链霉亲和素(Streptavidin-HRP)。洗涤后,加入显色底物TMB,避光显色。颜色反应的深浅与样本中IL-6Rα的浓度成正比。加入终止液终止反应,在450nm波长(参考波长570 - 630nm)测定吸光度值。 |

白细胞介素 6 受体是由 IL-6 受体亚基(IL-6Rα)和信号转导糖蛋白 130(gp130)组成的复合物。可溶性IL-6Rα的分子量约 50kDa,由膜 IL-6Rα经蛋白水解产生。该受体可在溶液中与 IL-6 结合,使 IL-6/IL-6sRα复合物与膜gp130 结合,增强IL-6 活性。目前调控体内可溶性 IL-6R 的脱落以及这些受体的功能和重要性尚未知。然而涉及 IL-6 水平升高的病理状态也许与可溶性 IL-6R 含量升高有关。
