自噬是凋亡之外的第二种程序性细胞死亡方式,是进化过程中保留下来的高度保守的自我保护机制,参与自噬的基因从单细胞的酵母到植物、脊椎动物和人中均较为保守。自噬在机体稳定、组织重塑、细胞发育及内环境稳态、生物合成以及环境的适应方面等都发挥着十分重要的作用。

根据细胞内底物运送到溶酶体腔方式的不同,哺乳动物细胞自噬主要可分为三种方式(下图):大自噬(Macroautophagy),微自噬(Microautophagy)和分子伴侣介导的自噬(Chaperone-mediated autophagy, CMA)。大自噬即我们说的自噬(autophagy)。微自噬是溶酶体直接内陷,包裹细胞质中的成分,形成一个内在的囊泡,没有形成自噬体的过程。分子伴侣介导的自噬为胞浆内蛋白结合到分子伴侣后转运到溶酶体腔中,被溶酶体酶消化。
