细胞毒性 T 淋巴细胞相关蛋白 4 (CTLA-4),又名 CD152,是免疫球蛋白超家族中的成员,在辅助型 T 细胞表面表达。 CTLA-4 类似于 T 细胞的共刺激蛋白 CD28,它们都结合 CD80 和 CD86。CTLA-4 传递抑制信号给 T 细胞,而 CD28 则传 递刺激信号。CTLA-4 可作为免疫检查点发挥功能、下调免疫系统、当结合抗原递呈细胞表面的 CD80 或 CD86 时作为“关 闭”开关。细胞内的 CTLA-4 同样存在于调节性 T 细胞中,对它们的功能发挥是重要的。该基因突变与胰岛素依赖型糖尿病、甲状腺机能亢进、桥本甲状腺炎、乳糜泻、系统性红斑狼疮、甲状腺相关眼病、 原发性胆汁性肝硬化和其它自身免疫性疾病有关。CTLA-4 基因的多态性与自身免疫性疾病如自身免疫性甲状腺疾病和多 发性硬化症有关,但这种关联性通常较弱。在系统性红斑狼疮中,人们发现可溶性 CTLA-4 的剪接变异体异常产生,且存 在于患有该病的患者血清中。
