ERBB3 分子靶点综述

人表皮生长因子受体3 (human epidermal growth factor receptor 3, Her3)，也称ErbB3，是一种I型跨膜糖蛋白，是酪氨酸激酶受体ErbB家族的成员。ErbB3结合神经调节蛋白NRG1和NRG2，但缺少一个有效的激酶结构域；与另一个ErbB受体的异二聚作用是信号转导所必需的。ErbB3在雪旺细胞、神经嵴细胞、心脏瓣膜和交感神经系统的发育中起重要作用。这种基因的扩增和/或其蛋白的过度表达在许多癌症中都有报道，包括前列腺癌、膀胱癌和乳腺癌。ErbB3在多种肿瘤中高表达，激活时可引起肿瘤增殖及药物抵抗性。E3泛素连接酶、单克隆抗体以及microRNA是当前调节ErbB3表达及功能的主要分子标志物。
