EGFR 分子靶点综述

表皮生长因子受体(EGFR),又名ErbB1和HER1,是一个I型跨膜糖蛋白。它存在于细胞表面,靠与它的特定配体包括EGF和TGF-α结合来激活。EGFR信号在调节细胞增殖、存活和分化过程中起重要作用。EGFR与配体结合后就会诱导其发生同源或异源二聚体化,进而引发后续的酪氨酸自磷酸化和多种下游通路(MAPK、PI3K/PKB和STAT)。该受体激活在人类皮肤的先天性免疫应答中有重要作用,EGFR过表达(上调)或活性过度增加与许多癌症,包括肺癌、肛门癌和多形性胶质母细胞瘤有关。异常的EGFR信号与牛皮癣、湿疹和动脉粥样硬化有关,目前,研究者认为EGFR可作为癌症治疗的一个靶标。
