PRLR 分子靶点综述

催乳素受体(PRLR)是细胞因子受体,由位于染色体5p13-14上的基因编码,可作为跨膜受体与催乳素相互作用。PRLR包括一个可结合催乳素的胞外区、一个跨膜区和一个胞质区。PRLR广泛在乳腺、胎盘、肾脏、肝脏和胰腺中表达,除了催乳素外,它还可被生长激素(GH)和人胎盘泌乳素(hPL)活化。研究表明PRLR的异常与某些肿瘤尤其是乳腺癌的发病及预后等密切关联。与PRLR特异性结合可刺激乳腺癌细胞生长,一些可与PRLR结合、但不使之形成二聚体的拮抗剂可抑制乳腺癌细胞生长。PRLR还存在几种异构体,其中可溶性的PRLR不能转导催乳素信号。
