NTRK1 分子靶点综述

TrkA(Tyrosine kinase receptor ,酪氨酸激酶受体A)是原癌基因Trk的产物,是神经营养酪氨酸激酶受体家族的成员,也被称为高亲和力神经生长因子受体1(NTRK1)。TrkA、TrkB和TrkC分别优先结合NGF、NT-4和BDNF、NT-3。所有Trk家族蛋白都有一个保守的复杂亚结构域组织,包括一个信号肽,两个富含半胱氨酸的结构域,三个富含亮氨酸的基序,细胞外区域的两个类似免疫球蛋白的结构域,以及一个包含酪氨酸激酶结构域的细胞内区域。两种不同的TrkA异构体因其胞外结构域的6个氨基酸插入而不同。较长的TrkA亚型是唯一在神经元组织中表达的亚型,而较短的TrkA主要在非神经元组织中表达。NGF以低亲和力与TrkA结合并激活其细胞质激酶,启动一个介导神经元存活和分化的信号级联。NGF的高亲和力结合需要TrkA与肿瘤坏死因子受体超家族成员之一的p75 NGF受体 (NGFR) 共表达。NGFR以低亲和力结合所有神经营养因子,调节Trk活性,并改变Trk受体对其配体的特异性。
