FGFR3 分子靶点综述

成纤维细胞生长因子受体3(FGFR3),又称CD333,是成纤维细胞生长因子受体(FGFR)家族成员,其氨基酸序列在家族成员间及不同物种间高度保守。FGFR家族成员在配体亲和性和组织分布上存在差异。FGFRs是具有细胞内酪氨酸激酶活性的跨膜催化受体。FGFR基因突变可导致多种以骨骼发育异常(如软骨发育不全)为特征的人类发育障碍,而FGFR表达上调可能导致细胞转化和癌症。
FGFR3作为全长代表性蛋白,其结构包含:由三个免疫球蛋白样结构域组成的胞外区、单个疏水性跨膜片段和胞内酪氨酸激酶结构域。FGFR3的胞外部分与成纤维细胞生长因子相互作用,启动下游信号级联反应,最终影响细胞有丝分裂和分化。FGFR3既能结合酸性也能结合碱性成纤维细胞生长激素,在骨骼发育和维持中发挥重要作用。FGFR3基因突变会导致颅缝早闭和多种类型的骨骼发育不良。目前已发现三种编码不同蛋白亚型的可变剪接转录变体。CD333是酸性和碱性成纤维细胞生长因子的受体。
