凝血因子VII(Coagulation factor VII),又称血清凝血酶原转化加速因子(Serum prothrombin conversion accelerator)、F7或FVII,是肽酶S1家族的成员。作为凝血级联反应的核心蛋白之一,它属于丝氨酸蛋白酶类酶,且凝血因子VII(FVII)缺乏症是最常见的罕见先天性出血性疾病。凝血因子VII包含两个EGF样结构域、一个Gla(γ-羧基谷氨酸)结构域和一个肽酶S1结构域。其主要作用是与组织因子(TF)共同启动凝血过程。组织因子位于血管外侧,通常不与血流接触。凝血因子VII的作用会受到组织因子途径抑制物(TFPI)的阻碍,后者在凝血启动后几乎立即释放。凝血因子VII依赖维生素K并在肝脏中合成。当血管受损时,组织因子暴露于血液和循环中的凝血因子VII。一旦与TF结合,FVII会被不同的蛋白酶激活为FVIIa,这些蛋白酶包括凝血酶(因子IIa)、因子Xa、IXa、XIIa以及FVIIa-TF复合物本身。
