CLU 分子靶点综述

丛生蛋白(Clusterin)是 1983 年首次由绵羊睾丸膜体液中分离得到的,是一种普遍存在的、高度保守的异源二聚体硫酸化糖蛋白,该蛋白广泛表达于机体多种组织中,发挥促细胞聚集、调节生殖、组织修复、转运脂类、补体抑制/调节免疫等多种功能。由于其具有广泛、未明机制的各种功能,因而具有多种命名,例如载脂蛋白 J、硫酸糖蛋白 2 (SGP-2)、TRPM-2 和 SP-40 等。在 1992 年英国剑桥召开的国际会议上将其统一命名为 Clusterin,简称 CLU。研究表明 Clusterin 不仅与多种生物过程相关,而且还与多种病理状态(如神经退行性疾病、衰老和癌症)有关。人 Clusterin 包含两个螺旋结构域、三个核定位信号域和一个肝素结合域。细胞内前体的裂解去除信号肽并产生相当大小的α链和β链,分泌为大约80kDa 的 N-糖基化和二硫键连接的异二聚体。
