ACP3 分子靶点综述
前列腺酸性磷酸酶(PAP/ACPP,亦称PSAP)是一种由前列腺产生的酶。作为一种非特异性磷酸单酯酶,前列腺酸性磷酸酶在雄激素的调控下被合成并分泌到精液中。该酶为二聚体结构,分子量约为100千道尔顿。作为前列腺癌的生物标志物,前列腺酸性磷酸酶在临床研究中具有重要意义;此外,它可能在受精过程中发挥一定作用。PAP的主要功能是利用其位于两个结构域之间的催化残基(His(12)和Asp(258))来使大分子去磷酸化。细胞内的前列腺酸性磷酸酶属于真正的酪氨酸磷酸酶,被认为可通过使ErbB-2去磷酸化来抑制前列腺癌细胞的生长。在前列腺癌细胞中,这种酶发挥中性蛋白酪氨酸磷酸酶的作用,能将HER-2/ErbB-2/Neu分子上的磷酸酪氨酸残基去磷酸化。将PAP的分泌型异构体注射到慢性疼痛小鼠模型体内可产生显著的镇痛效果。该酶还具有外切5’-核苷酸酶活性,分布于体内多种组织,并与慢性疼痛的形成有关。另外,对于患有非前列腺腺癌(如结肠癌和胃癌)的患者而言,PAP可能成为特定免疫疗法的靶标分子。
