KMT2A 分子靶点综述

硫氧还蛋白(Trxs)是一群广泛存在于所有活细胞中的小分子蛋白,是细胞氧化还原平衡的关键调节者。哺乳动物的Trx家族有三个成员:Trx-1、Trx-2和TrxL。Trx-1是唯一一个存在于细胞外的硫氧还蛋白,由淋巴细胞、肝细胞、成纤维细胞和多种肿瘤细胞分泌产生。血浆中Trx-1的浓度可达到6 nM。它主要存在于细胞的细胞质中,少量存在于细胞核以及与细胞外表面相关的地方。在各种应激条件下如缺氧、过氧化氢浓度升高、光化学氧化应激、病毒和细菌感染,Trx-1的表达增加。Trx-1的生物学功能包括生长因子活性、抗氧化性能、提供还原当量的辅因子和转录调控。类风湿性关节炎患者的滑膜组织在氧化应激条件下,其Trx-1水平增加。血浆中Trx-1水平与疾病的严重程度有关,表明Trx-1可作为病理条件的生物标志物。
