EPCAM 分子靶点综述

上皮细胞黏附分子(EpCAM),又称CD326、GA733-2抗原等,是一种I型跨膜糖蛋白。其结构包含:具有两个EGF样重复序列和半胱氨酸富集区的胞外域、跨膜域以及胞质域。该分子不仅调控细胞黏附和增殖,其过表达现象已在多种上皮源性肿瘤中被发现,并与某些肿瘤的高分期、高级别及不良预后显著相关。
研究表明,EpCAM作为一种钙离子非依赖性的同嗜性细胞黏附分子,其作用机制与已知的四大细胞黏附分子超家族均无明确关联。研究进展揭示,EpCAM除参与细胞黏附外,还在细胞增殖、迁移及分化过程中发挥重要作用。值得注意的是,近期研究发现EpCAM可作为Wnt/β-catenin信号通路的靶基因,具有潜在的预后评估价值。
该分子具有致癌潜能:当其胞内结构域被释放后,可通过激活Wnt信号通路的关键元件向细胞核转导信号。这一机制为理解EpCAM在肿瘤发生发展中的作用提供了新的分子基础。
