【DESCREPTION】Interleukin 6 (IL-6) is an interleukin that acts as both a pro-inflammatory cytokine and an anti-inflammatory myokine. In humans, it is encoded by the IL-6 gene. IL-6 is secreted by T cells and macrophages to stimulate immune response, e.g. during infection and after trauma, especially burns or other tissue damage leading to inflammation. IL-6 also plays a role in fighting infection, as IL-6 has been shown in mice to be required for resistance against bacterium Streptococcus pneumoniae.
【PRINCIPLE OF THE ASSAY】This assay employs the quantitative sandwich enzyme immunoassay technique. A monoclonal antibody specific for rat IL-6 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and IL-6 present is bound by the immobilized antibody. After washing away any unbound substances, a biotin-linked detect antibody specific for IL-6 is added to the wells. Following a wash to remove any unbound antibody-biotin reagent, streptavidin-HRP is added. After washing, amplification reagent is added to the wells. Following incubation any unbound substances is removed during a wash step and streptavidin-HRP is added. After washing, substrate solution is added to the wells and color develops in proportion to the amount of IL-6 bound in the initial step. The color development is stopped and the intensity of the color is measured.
